Based on the TV Series of the same name, insects all over the world begin to swarm for unknown reasons, with other animals and plants acting strange as well. To investigate this, Takuya Kai is dispatched with an Earth Academia group. Investigating independently, Takuya finds Guru who reveals that insects are preparing for battle against invaders from another dimension. Offering their aid, the Earth Academia create suits of armor to combat the threat as the Jamahl arrive to Earth to enslave humans. Though they have trouble perfecting the Bio-Machinery, Guru arrives to infuse the armor with the life force of many insects. Once the suits become the B-Commanders, one suit chooses Takuya while the other two bond to Daisaku Katagiri and Rei Hayama. In the future, Earth has become so polluted that humanity is forced to live in enclosed shelters. In Japan, the shelters are divided between the "inside", which caters to the country's lucrative anime industry, and the "outside", where most people work to support the people working on the "inside". S-zawa is one such "outside" worker whose job involves sending messages into space. One day, he's approached by the ailing embodiment of Earth to send a message before she collapses. Shortly after the message is sent, other girls similar to Earth start showing up at S-zawa's workplace in an effort to help Earth, but he has a thing or two to learn about what the planets and their moons are really like.

The plot revolves around an intranet-based online game (D.P. = Danger Planet) which is played using V.P. (Virtual Puppet). VP is literally an avatar which a person uses to in the game. However, unlike avatars (characters) from current online games, a VP is allowed to be modified (modded) by a person if he or she has sufficient programming skills.

UFO Robot Grendizer is the third entry in the Mazinger Trilogy by Go Nagai, following Mazinger Z and Great Mazinger.
